Oral Antivirals: HCV Protease and Polymerase InhibitorsThe NS3-4A serine protease enzyme and the NS5B RNA-dependent RNA polymerase enzyme are primary targets for oral antiviral agents, since both enzymes are essential for HCV replication. Unfortunately, HCV's high replication rate -- billions of copies... Read more »FDA Review of Viral RNA TestingShortly after the approval of the three protease inhibitors, the FDA reviewed the diagnostic assays that quantitate HIV RNA.These tests are being used by many clinicians to monitor viral loads in their patients and to assess their response to... Read more »"Smart Bomb" RNA Molecule Could Take Out Drug-Resistant HIVResearchers have engineered an RNA molecule to block freely circulating HIV as well as replication inside infected cells. Tests in mice engineered to be susceptible to HIV showed the chimeric RNAs reduced HIV-1 replication by several orders and... Read more »

Using Polymerase to Combat Infection?I have been told that there aren't many t-4 receptors in the human body. If the genetic code for ~only~ the t-4 receptor could be isolated, couldn't PCR replicate the receptor? What I'm wondering is this -- couldn't infected...Response from Rick Stryker, M.D., M.P.H.
University of California Los Angeles School of Medicine
Interesting thought, that perhaps one could "mimic" the process of HIV binding to T-4 (AKA CD4+ or helper cells)and trick the virus into releasing its deadly package of instructions (RNA) harmlessly into some non-infectious state....

HIV RNA Test/HIV DNA PCR TestDr Brook, to one of your answers dated 28 October 2001, DNA PCR Test while Pregnant you said that the HIV RNA test is more sensitive than the HIV DNA PCR test for detecting early infection if you are not taking antiretrovirals. Now does this mean...Response from Brooks Jackson, M.D.
Johns Hopkins University, School of Medicine
Once an adult individual is infected with HIV and is antibody positive, the use of antiretrovirals for treatment will not cause the ELISA test for HIV antibody to become negative. BJ...

Does anybody know the difference between RNA-PCR and RNA-bDNA tests? Are these tests accurate? Any false negative or false positive results? Are they worth taking to evaluate early HIV infection when possible early HIV infection symptoms are present? I... Read more » < 400 copies of HIV RNA per 1 ml of plasma (????)
