Safety and Efficacy of Once-Daily Dose of Didanosine (DDI)
September 1997 Dr. E. Martinez and colleagues from Barcelona, Spain, reported on 80
patients who received ddI once a day, plus D4T 40 mg bid, with or without
saquinavir or indinavir. The mean baseline CD4 count was 178 cells/mm3, and
the mean plasma viral load was 4.9 logs. Less than 10% of the patients
complained about gi side effects or had to withdraw from the study. CD4
counts increased a mean of 86 cells/mm3 and viral load decreased 1.9 logs
at 3 months. 43% of patients had peripheral viral loads below 200 copies/ml
(all of these were taking a protease inhibitor). The data suggest that
daily dosing of ddI is consistent with a good clinical response to
combination chemotherapy and deserves further study.
